Net Charge and Oxygen Affinity of Human Hemoglobin Are Independent of Hemoglobin Concentration

چکیده
I N T R O D U C T I O N Gary-Bobo and Solomon (1) have proposed that the anomalous osmotic behavior of the erythrocyte is due to a decrease of the net charge of hemoglobin which occurs when the hemoglobin concentration inside the erythrocyte is raised. Employing Donnan equilibrium studies, they subsequently (2) produced evidence to show that a hemoglobin charge dependence on hemoglobin concentration also exists in vitro. This phenomenon, which has been interpreted by GaryBobo and Solomon (1, 2) in terms of aggregation of hemoglobin molecules, is often quoted as an example of altered functional properties of hemoglobin at high concentrations (3, 4), and has been the subject of two Letters to the Editor in this Journal (5, 6). During recent studies on the diffusivity of hemoglobin, whose dependence on hemoglobin concentration increases sharply above a concentration of 30 g/dl (7), we became interested in the behavior of other functional properties o f hemoglobin when the hemoglobin concentration rises up to and above the values found inside erythrocytes. Net charge and oxygen affinity of human hemoglobin are independent of hemoglobin concentration
The dependence of net charge and oxygen affinity of human hemoglobin upon hemoglobin concentration was reinvestigated. In contrast to earlier reports from various laboratories, both functional properties of hemoglobin were found to be independent of hemoglobin concentration. Two findings indicate a concentration-independent net charge of carbonmonoxy hemoglobin at pH 6.6: (A) The pH value of a ...
متن کاملNew Sequential Model for Human Hemoglobin: Alpha Subunit as Cooperativity Inducer
Hemoglobin is a tetrameric oxygen transport protein in animal bodies. However, there is a paucity of information regarding differences between alpha and beta subunits of hemoglobin in terms of oxygen affinity. The sequential model of Koshland, Nemthy and Filmer (KNF model) has attributed similar affinities to both alpha and beta subunits. The main purpose of the present study is to construct a ...
متن کاملA tri state mechanism for oxygen release in fish hemoglobin: Using Barbus sharpeyi as a model
Hemoglobin is a porphyrin containing protein with an a2b2 tetrameric structure and like other porphyrin compounds shows spectral behavior of species specific characteristics. Researchers tend to relate bands in the hemoglobin spectra to certain structural and/or functional features. Given the fact that hemoglobin is the main oxygen carrier in animals functioning through the Oxy«Deoxy equilibriu...
متن کاملChange in Oxygen Absorption of Human Adult and Fetal Hemoglobin Due to 940 MHz Electromagnetic Field Radiation Exposure
The effects of electromagnetic fields (EMFs) radiation at the frequency of 940 MHz on the structure and function of human adult and fetal hemoglobin (HbA and HbF) were studied. After extraction and purification of HbA and HbF, the oxygen absorption values for exposed and unexposed HbA and HbF to EMF were compared. The slope of oxygen absorption curve for exposed HbA was increased while that for...
متن کاملSolvent regulation of oxygen affinity in hemoglobin. Sensitivity of bovine hemoglobin to chloride ions.
Under physiological conditions of pH (7.4) and chloride concentration (0.15 M), the oxygen affinity of bovine hemoglobin is substantially lower than that of human hemoglobin. Also, the Bohr effect is much more pronounced in bovine hemoglobin. Numerical simulations indicate that both phenomena can be explained by a larger preferential binding of chloride ions to deoxyhemoglobin in the bovine sys...
